CPB-mediated increases in fibronectin matrix assembly by cultured fibroblasts. A, a representative blot from a single experiment shows levels of biotinylated FN (top) incorporated by fibroblast monolayers into deoxycholate-insoluble matrix for samples normalized to vimentin (bottom). Cell layers were treated for 24 h with complete medium supplemented with biotinylated FN (20 μg/ml) containing 25 mm HEPES (pH 7.1) with or without anastellin (150 μm), CPB (50, 250, and 500 μm), or CPB(W6A) (500 μm) prior to extraction in 2% deoxycholate. B, biotinylated FN matrix incorporation assessed by quantifying the ratio of band densities for biotinylated FN and vimentin within each sample. Data are pooled from three independent experiments. Addition of 150 μm anastellin or 250 μm CPB to IMR90 cells significantly increased assembly of biotinylated FN into the matrix as compared with untreated cells (*, p < 0.05, two-tailed). Cells treated with high concentrations CPB (250 and 500 μm) showed similar levels of FN matrix incorporation as 150 μm anastellin (p > 0.05, two-tailed), unlike cells treated with 50 μm CPB or 500 μm CPB(W6A) (*, p < 0.05, two-tailed).
