Four routes of modification of compound 10 to obtain trifucosylated core structures. Alkylamine-modified 10 was core α1,6-fucosylated (with FUT-8) and then modified by FUT-1, FUT-6, β-hexosaminidase, and GALT-1 in various serial reactions (A–D); reactions were monitored by MALDI-TOF MS. Quasimolecular ions in the mass spectra are [M + Na]⁺; transfer of fucose or galactose is indicated by respective gain of 146 or 162 mass units and removal of N-acetylglucosamine by loss of 203 mass units.
