HCV RLuc cells were transfected with the ORP4 expression plasmid. pcDNA3 was used to equalize the total amount of plasmid DNA among the transfections, and pCMV-βGal was included in each transfection to normalize the transfection variations. The transfected cells were cultured for 48 hr and then treated with lipid synthesis inhibitors cerulenin (CRLN) (A), or lipid sequestering agent methyl-β-cyclodextrin (MβCD) (B) for 24 hr. Cell lysates were prepared for the β-Gal activity and luciferase activity assay. The data are Means ± SD of triplicates and are representative of at least three independent experiments.
