Measurement of H3K27me3 level in SU-DHL-6 cells using Western blot following treatment for 3 days at 37°C in 5% CO₂. The concentration response of reference inhibitors DZNep (A) and RBC124 (B) in H3K27me3 Western blot cell-based assay. Total histone H3 is shown as a loading control. Data for the modified and total histone were quantitated using the Gel-Pro Analyzer. Evaluation of the cellular inhibitory activity of the reference inhibitors generated an IC₅₀ value of 7.4 and 0.16 μM for RBC124 (C) and DZNep (D), respectively.
