TTX does not eliminate I_NS and Ni²⁺ abolishes I_Ca component. In A, during the 260 mV sec⁻¹ ramp, I_Na1 was superimposed on I_NS which was followed by the I_Ca component (shaded area). TTX (30 μmol L⁻¹, B) eliminated I_Na1 (short arrow) but not I_NS as confirmed by the presence of I_PS. Ni²⁺ (2 mmol L⁻¹, C) abolished the I_Ca component (long arrow). Recovery is shown in D. In inset 1, depolarizing steps from V_h −40 mV to +20 mV elicited I_Ca (control, E) which was not suppressed by TTX (TTX, F) but was eliminated by Ni²⁺ (TTX + Ni²⁺, G). During the recovery from TTX and Ni²⁺, I_Ca returned to control value (recovery, H).
