FAD and FMN content measurement: Protein-bound FAD and FMN content were measured using HPLC. Flavins were extracted from 50 μM of wt or arginine mutant preparations of both nNOSred and iNOSred by boiling and pelleting the precipitated protein by centrifugation. The supernatant, which contains the extracted flavins, was analyzed using HPLC (Experimental Procedures).
