Gln3-Myc¹³ localization and G₁ arrest of wild-type cells following the addition of α-factor are not affected in parallel. Wild-type cells were pregrown in YNB-glutamine medium and sampled at the indicated times. The bud index (A) or Gln3-Myc¹³ localization (B and C) were then determined. The time at which cells G₁-arrested (greatest percentage of unbudded cells) was determined as in Fig. 3, B and C. α-factor (2 μgm/ml) was added to the culture immediately after the zero and 2.0 h time point samples were taken (arrows).
