The gle2-patch mutant is able to transport mRNAs to the cytoplasm under mild ethanol stress. The gle2-1 and gle2-gis strains, but not the gle2-patch mutant strain, are impaired in poly-(A)⁺ RNA export after ethanol shock. Indicated strains were grown at 30°C till mid log phase and treated with 5% ethanol (v/v) for 5 min (left). Strains without ethanol treatment served as negative controls (right). Localization of poly-(A)⁺ RNA in the indicated strains was assessed by fluorescence in situ hybridization using a Cy3-oligo(dT)³⁰ probe. Nuclear and mitochondrial DNA was stained with DAPI.
