MEKK2 regulation of IL-6 expression. A, MEKK2^−/− mouse embryonic fibroblasts were infected with retrovirus containing empty vector, FLAG-WT MEKK2, or FLAG-T283A MEKK2 cDNA. Following 5 days of selection in puromycin, resistant pools were isolated and immunoblotted for expression of MEKK2. B, stable puromycin-resistant pools of MEKK2^−/− MEFs expressing various forms of MEKK2 were plated in triplicate in 24-well dishes at 30,000 cells/well and then treated with TNFα (1 ng/ml) for 18 h. Supernatants were harvested, and IL-6 was quantified using the Quantikine immunoassay kit (R&D Systems). Error bars represent S.D.
