Plasmid pRG24 carries the 5′ region of the cotH coding region posed under the transcriptional and translational control of the cotA promoter. The non-replicative plasmid was used to transform competent cells of the wild type strain PY79. Chloramphenicol-resistant clones originated by a single cross-over event between homologous DNA regions on the plasmid and on the chromosome. In the resulting strain, RG24, the cotH promoter (PH) transcribes a truncated form of cotH while an entire gene is transcribed by the cotA promoter (PAH).
