NHFs were transfected with pShooter plasmid DNA carrying human SEPP1 cDNA (SEPP1) [23]. NHFs transfected with plasmid DNA without an insert sequence (empty) were used as a control. (A) SEPP1 expression was assayed by measuring both mRNA and secreted protein (inset) levels; (B) DHE oxidation at 48 h post-irradiation; cell survival was assessed by measuring (C) the percentage of PI-positive (nonviable) cells, and (D) a clonogenic assay. Representative dishes with colonies are shown in (D) for comparison. Error bars represent standard deviation of the mean.
