Representative gel image from the independent experiment using IPG 3–10NL strips. This independent experiment validated the data acquired from gels with narrow pH range. Ten protein spots were identified to be differentially expressed (P<0.05) between anorexic tumour-bearing mice (n=5) and free-fed mice (n=5). The expression pattern of these proteins (marked in red) was identical to that in the gels with narrow pH range. Data for the six other proteins that were identified in gels using IPG 3–6 and IPG 5–8 strips could not be validated as they were not visible in the gels with wide pH range owing to much lower resolution capacity of IPG 3–10NL strips. Proteins of hypothalamus (150 μg) were resolved in 13% SDS–PAGE gels in the second dimension and stained with SYPRO Ruby.
