IGFBP-6 does not directly phosphorylate PHB2 in vitro. Recombinant PHB2 (5 μg) was incubated with WT or mIGFBP-6 (5 μg) in the presence or absence of 100 μm ATP. Samples were analyzed by phosphotyrosine (pY) Western blotting. Phosphorylation of N-ezrin (5 μg) by the EGF receptor (EGFR; 1.62 units) was used as a positive control.
