The NEG domain of p53 contains the lysine residues targeted by HDM2 for ubiquitination and degradation of p53. Domain study of p53 reveals that the NEG domain is essential for the association between HEXIM1 and p53 [113]. It is proposed that HEXIM1 binds and stabilizes p53 protein by blocking the ubiquitination of p53 by HDM2.
