ScsC is required for S. Typhimurium copper resistance in vitro. (A) Disk diffusion assays of wild-type S. Typhimurium SL1344, SL1344scsC and SL1344scsC complemented with plasmid pScs. Sterile filter disks were placed on bacterial lawns and treated with 5 μl CuSO4 (1 M). The data presented are representative of five independent repeats. (B) Copper sensitivity and complementation of S. Typhimurium SL1344 scsC mutant was tested by spotting a series of 10-fold dilutions of cultures of the same density onto Tris agar plates containing increasing amounts of copper. Pictures were taken after a 24-h incubation at 37°C in the presence (right panel) or absence (left panel) of 2.8 mM CuSO4.
