DNMT1 Flashplate™ SPA principle. Biotin is represented as a gray circle, tritium atoms as stars and the lightning represents the ability of a tritium atom to excite the plastic scintillator. In homogeneous phase, the hemimethylated duplex is methylated by the studied DNMT. The biotin is then captured and the methylation reaction is stopped on a streptavidine-coated Flashplate™. Only the bound 3H-CH3-DNA can excite the coated scintillant and emit a signal.
