A, Oxidized (dashed line) and reduced (solid line) UV-Vis spectra of LmCytc. The maxima used to follow oxidation of reduced LmCytc by LmP is at 558 nm. B, Determination of Δε558_red enzymatically. Aliquots of 2.2 μM H₂O₂ was added until LmCytc could no longer be oxidized by LmP. The ε558 of the enzymatically oxidized sample was determined to be 9.6 mM⁻¹•cm⁻¹ so that Δε558_red was calculated to be 19.4 mM⁻¹•cm⁻¹. The sample was then fully reduced by addition of granules of sodium dithionite.
