Selectivity of N-aryl piperazines across other mGlu subtypes. A single concentration (10 µM) of VU0364289 (closed squares) and DPFE (open squares) was assessed for modulation of orthosteric agonist activity (closed circles) (glutamate at all except mGlu₇ where L-AP4 was used) across the other seven mGlu subtypes. Activity at mGlu₁ was investigated using fluo-4 fluorescence-based measurement of Ca²⁺ mobilization. Activity at all other subtypes was assessed by a fluorescence-based assay of thallium-flux through GIRK channels (Niswender et al., 2008). Data represent the mean ± S.E.M. of three independent experiments performed in triplicate, with the exception of mGlu₁ where the mean of two independent experiments are shown.
