S195D enhances, whereas wild-type RNPC1 and S195A inhibit, eIF4E binding to p53 mRNA. HCT116 cells were uninduced or induced to express HA-tagged wild-type RNPC1 (A), S195D (B), and S195A (C) for 48 h followed by immunoprecipitation with a control IgG or anti-HA that recognizes RNPC1 or anti-eIF4E. Total RNAs were purified from immunocomplexes and subjected to RT–PCR analysis to measure the level of p53 and actin mRNA. The relative level of p53 mRNA was measured by densitometry, and the relative fold change is shown below each pair.
