Transcriptional regulation of K15 keratin gene expression by the C/EBP transcription factors. (A) Cotransfection experiments. K15 wt-CAT was cotransfected into HeLa cells with constructs expressing C/EBP-β or CHOP. The basal activity of the K15 wt-CAT construct was considered to be 1, and the relative activities of the promoter in the presence of cotransfected constructs are shown. β-Galactosidase activity was used to normalize the CAT activities. (B) Binding of C/EBP-β to the K15 promoter region by gel shift analysis with the purified CEB/Pβ protein and the −266/−353 probe. Arrows point to the bound C/EBP-β protein on the DNA.
