Comparison of nonenzymatic primer extension reactions on homopolymeric A4 and G4 templates using T and 2-thio-T monomers. (A) Chemical structure of monomer 3′-NH2-2-MeImpddsT. The reaction was initiated by addition of 10.0 mM 3′-NH2-2-MeImpddsT or 3′-NH2-2-MeImpddT. Arrows indicate primer and full-length product. Primer extension is faster using 2-thio-T vs. T on the A4 template, but slower on the G4 template.
