Macrophages were pretreated with PD98059 (P, 50 μM) (A, C) or anti-TLR2, anti-TLR4 or anti-Dectin-1 antibodies (B) for 1 h; the cells were incubated with β-glu6 (G, 100 μg/mL). The levels of ERK 1/2 (both total and phosphorylated), phosphorylated c-Raf and beta-actin were detected by western blot assay. Real-time PCR was used to examine IL-6 and IL-12 mRNA expression levels in macrophages incubated with β-glu6 (G, 100 μg/mL) for 4 h (C).
