Figure 4

Mapping of the crucial region of SKP1Maize involved in the P7-2-SKP1Maize interaction. (A) Two types of SKP1 proteins. Both of the two types have two conserved domains (Skp1_POZ and Skp1) and two variable regions. Type II proteins have an additional C-terminal region. (B) Schematic representation of SKP1Maize and its truncations in the study. The full-length SKP1Maize (spanning residues 1 to 176) and its truncations are indicated by pink bars while the deleted regions by dashed lines. The numbers denote the amino acid positions of the proteins. The ability of different SKP1Maize truncations to interact with P7-2 is indicated on the right (+, positive; -, negative). (C) The interaction between P7-2 and different SKP1 truncations in YTH assays. Yeast colonies expressing AD-SKP197-176 with BD-P7-2 were able to grow on the selective medium and turned slight blue. No growth was observed in the yeast expressing BD-P7-2 with AD-SKP11-67, AD-SKP11-101 or AD-SKP11-128.
