Thrombin does not modulate PAR1 or PAR4 homodimers. HEK293 cells were transfected with HA-PAR1-Luc (0.5 μg) and V5-PAR1-GFP (0–2.5 μg) (A) or HA-PAR4-Luc (0.03 μg) and V5-PAR4-GFP (0–0.24 μg) (B). Forty-eight hours post-transfection, the cells were unstimulated or treated with α-thrombin (10 nm) and analyzed for GFP expression, luciferase activity, and BRET. The data are from three independent experiments in which all points were analyzed by global fit to hyperbolic or linear curve.
