Effect of mutations at Gln163. (A) The Gln163Lys and Gln163Asp mutations abolish the response to Ca2+. (B) The Gln163Met mutant retains a response to Ca2+. (C) In the X-ray structure of mCherry (PDB ID 2H5Q) [24], Gln163 is directed into the interior of the protein in close proximity to the chromophore. Compare the orientation of Gln163 with the orientation of Lys163 in R-GECO1 as shown in Figure 1B.
