(B) Serum-starved A375 cells were treated with 100 ng/ml rhPTN and harvested at various time-points. The activation of ERK1/2 was detected by Western blotting. (C) A375 cell lines were treated using LY294002, a pI3K inhibitor 48 hrs, and cell proliferation was measured by MTT. (D) A375 cell lines were treated with U0126 at 0.1, 1 and 10 μM, a MEK1/2 inhibitor 48 hrs and cell proliferation was measured by MTT. (E) A375 cells treated with LY294002 were added to upper filter and cell migration was determined. (F) A375 cells treated with U0126 were added to upper filter and cell migration was determined.
