Induction of the unfolded protein response by chrysin. (A) HepG2 and SMMC-7721 cells were treated with chrysin at the indicated dosages for 24 hrs. Cell lysates were harvested and subjected to Western blot analysis with anti-GRP78 antibody. β-actin was probed as a loading control. (B) HepG2 cells were treated with 20 μM chrysin for indicated periods. Total proteins were harvested and subjected to Western blot analysis of the phosphorylated eIF2α. (C) HepG2 cells were treated with 20 μM chrysin for indicated periods. Total proteins were harvested and subjected to Western blot analysis of the spliced XBP-1 (XBP-1s). (D) HepG2 and SMMC-7721 cells were treated with indicated doses of chrysin for 24 hrs. Total proteins were harvested and subjected to Western blot analysis of cleaved caspase-7.
