miR-30e overexpression stimulated adipocyte formation and blocked osteoblast differentiation from marrow stromal cells. The miR-30e-expressing lentivirus increased mature miR-30e in primarily cultured mouse marrow stromal cells (a). In the presence of AIM, miR-30e-expressing lentivirus promoted adipocyte formation (b and c), increased mRNA levels of PPARγ, C/EBPα, C/EBPβ and aP2 (d) and induced protein levels of PPARγ, C/EBPα and aP2 (e). In the presence of OIM, miR-30e overexpression blunted ALP staining (f), decreased mRNA levels of Runx2, osterix, osteocalcin, ALP and BSP (g) and reduced protein levels of Runx2, osterix and ALP (h).
