LRP6 negatively regulated adipogenesis. The infection of the LRP6 RNAi adenovirus downregulated the endogenous LRP6 mRNA (a) and decreased β-catenin protein in 3T3-L1 cells (b). The luciferase activity of the β-catenin/TCF reporter was reduced following knockdown of LRP6 in 3T3-L1 cells (c). In the presence of AIM, the knockdown of LRP6 increased adipocyte formation (d and e), induced the mRNA levels of PPARγ, C/EBPα, C/EBPβ and aP2 at 48 h (f) and the protein levels of PPARγ, C/EBPα and aP2 at 72 h (g).
