Influence of nemorosone on cell cycle regulation in LAN-1. A: Cell cycle distribution of LAN-1 cells after exposure to 8 μM of nemorosone for 24 hrs. Cytometric analyses were performed using propidium iodide (PI) staining and BrdU incorporation. A highly significant increase of cells in sub-G0/G1 and G0/G1 (n = 3, P < 0.001)** phases as well as a decrease of those in S-phase (n = 3, P < 0.001)** is shown. No influence of nemorosone in the G2/M was observed (n = 3, P < 0.631). Results are represented as mean ± standard deviation of an experiment performed in triplicate. B: Influence of nemorosone on the protein level of p21Cip1 in LAN-1 as detected by immunoblotting. Induction of p21Cip1 after exposure to nemorosone for 24 hrs. Lane 1: control, lanes 2–4 increasing concentrations of the drug. An induction of this Cyclin-dependent Kinase Inhibitor (CDKI) is detectable after incubation with 1-fold the IC50 of nemorosone. The results depicted are representative for three independent experiments.
