Panel A) MCF-7 cells were transfected with plasmid DNA encoding PTEN(WT), PTEN(C124S), PTEN(G129E), PTEN(399stop) or the empty vector pEGFP-C2. The mean number colonies present after selection in 2 mg/ml G418 for approximately 3 weeks and corresponding standard deviation was calculated from 9 replicate wells for each plasmid. Asterisks indicate statistically significant decreases in the number of colonies present in comparison to pEGFP-C2 transfected cells (***, P<0.001).
