A. Real time RT-PCR showed that U0126 did not decrease NIS mRNA levels in MCF-7-tRA/H cells. MCF-7 cells treated with tRA/H were cultured in the presence of DMSO or U0126 for 24 hours. NIS mRNA level was normalized with GAPDH and the values of NIS/GAPDH mRNA ratio of MCF-7 cells without treatment were arbitrarily assigned. The results are representative of three independent experiments performed in triplicate and the mean ± SD are shown. B. Real time RT-PCR showed that U0126 did not significantly decrease NIS mRNA levels in cytosolic fraction in MCF-7-tRA/H cells. Cytosolic NIS mRNA level was normalized with GAPDH and the values of cytosolic NIS/GAPDH mRNA ratio of MCF-7-tRA/H cells were arbitrarily assigned. The results are representative of two independent experiments performed in triplicate and the mean ± SD are shown.
