Silica coating of mouse islets does not alter transcript levels of β-cell-associated genes. Real-time PCR analysis indicates that encapsulated islets retain gene expression levels, consistent with those of nonencapsulated controls. Quantification of gene regulation for (A) Actb (cytoskeletal element), (B) Pdx1 (β-cell-specific gene regulator), (C) pre-Ins2 (insulin gene pre-mRNA), and (D) Ins1/2 (gene encoding insulin) was conducted. All data are presented as fold up/downregulation of transcripts of respective genes relative to the Tbp housekeeping gene. *Significant (P ≤ 0.05) relative to nonencapsulated controls at the respective time point. All sample pairs were analyzed by one-way ANOVA. Error bars, ±1 SE (n = 3–10 replicates per condition).
