Adult OLPs do not generate astrocytes. Sections of P45 + 90 Pdgfra-creERT2/Rosa26-YFP mice were immunolabeled for YFP and GFAP. (a–d) No double-labeled cells were observed anywhere in the corpus callosum or cortex. Sections were also immunolabeled for YFP and S100β, a marker of protoplasmic astrocytes and some oligodendrocyte lineage cells. (e) In cortex, some YFP+ S100β+ cells were present, but these invariably colabeled for PDGFRA (yellow arrow), identifying them as OLPs. (f) In the corpus callosum, all of the YFP+ S100β+ cells were SOX10+ (yellow arrow in f). The white arrow in f indicates a YFP−,SOX10+ S100β+ oligodendrocyte lineage cell and the white arrowhead indicates a YFP− SOX10−,S100β+ presumptive astrocyte.
