CDK10 is a cyclin M-dependent protein kinase. (A) In vitro protein kinase assay on histone H1. Lysates from insect cells expressing different proteins were purified on a glutathione Sepharose matrix to capture GST-CDK10(wt or kd) fusion proteins alone, or in complex with STR-CycM fusion protein. Purified protein expression levels were analyzed by Western blots (Top and Upper Middle). The kinase activity was determined by autoradiography of histone H1, whose added amounts were visualized by Coomassie staining (Lower Middle and Bottom). (B) Same as in A, using purified recombinant 6His-ETS2 as a substrate.
