(a) Size exclusion chromatogram of 5P12-RANTES-E66S with (dashed line) and without (solid line) 0.5% FC-12 performed on a Superdex 75 10/300 GL column (GE Healthcare) in 50 mM HCOOH pH 3.8 at room temperature and 0.5 mL/min flow rate. The large decrease of the retention volume (from 16.2 to 10.7 mL) resulting from a major increase of mass (to ≥36 kDa) suggests a direct interaction between 5P12-RANTES-E66S and FC-12. (b) pH-dependent effect of detergent on RANTES-E66S secondary structure measured by CD spectroscopy at 25°C. The spectrum of RANTES-E66S at pH 3.8 without detergent (solid line) changes dramatically after addition of 0.5% FC-12 (dashed line). However, at pH 7.7, the spectra without detergent (dotted line) and after addition of 0.5% FC-12 (dashed-dotted line) are very similar.
