Overview of steps from drug administration to analysis involved in bioorthogonal proteomics. Live cells are incubated with a TCO-drug conjugate. Cell lysates are then prepared and TCO-drug bound to target protein is isolated using a Tz-labeled cleavable linker decorated on streptavidin magnetic beads. Protein is then run on an SDS-PAGE gel and desired bands are isolated and submitted for mass spectrometry analysis.
