Effects of Genistein on BMP2/SMAD5/RUNX2 transcription and downstream gene expression in cultured hBMSCs. Real-time RT-PCR analyses revealed that five representative genes (A) BMP2, (B) SMAD5, (C), RUNX2, (D), ALP, and (E) OSTEOCALCIN (OSC) were differentially expressed in Genistein (GEN, 10-6 M)-treated and vehicle control hBMSC cultures in the absence or presence of a complete estrogen receptor (ER) antagonist, ICI182780 (ICI, 10-7 M). Consistent with the results of microarray analysis, normalized transcript levels of BMP2, SMAD5, RUNX2, ALP, and OSC are shown to be upregulated in GEN-treated cells, and these five genes have significant difference between GEN-treated and the vehicle control group. In addition, ER antagonist ICI completely abolished the GEN-induced osteogenic gene transcription in cultured hBMSCs. Data are mean ± S.D. from triple independent experiments.
