Sphere-forming assay revealed that the Lin28 stably expressed MCF-7 clones pc-Lin28-1 and pc-Lin28-2 displayed an increased mammosphere formation rate when compared with empty vector and Lin28-mut transfectants (A). Meanwhile, the mammashpere formation rate was remarkably decreased in MDA-MB-231 cells that had been transfected with Lin28-siRNA (B). C. A soft-agar assay revealed that colony formation decreased in MDA-MB-231 cells after transfection with Lin28-siRNAs. D and E. By FCM, pc-Lin28-1 cells showed a 3-fold increase of ALDH1 activity as compared with empty vector cells (D).
