(A) JURL-MK2 and SUP-B15 cells were treated with 100 nM nilotinib or 2 μM BEZ235 for 24 h. MDM2 protein levels were analyzed by Western blot. GAPDH levels served as loading control. (B) MDM2 protein expression in SUP-B15 cells was tested after 6 or 24 h with different concentrations of nilotinib and BEZ235. (C) JURL-MK2 and SUP-B15 cells were treated with various doses of nilotinib or BEZ235 as indicated for 6 and 24 h. mTOR and its downstream targets, S6 and 4E-BP1, together with their phosphorylation forms were determined by Western blot.
