The kinetics or extent of TCR signaling is altered in CD4 hAPBTs that lack FAK. A, CD4 hAPBTs were transduced with a lentivirus containing miRNAs against Luciferase (Luc) or FAK. The cells were then stimulated by soluble anti-CD3 and anti-CD4 cross-linking for various times. The site-specific phosphorylation of LAT Y191, SLP-76 Y128, and AKT S473 was analyzed by immunoblotting. The expression of FAK, Pyk2, and actin was also examined.
