Pet colocalizes with CK8 and CK10 on epithelial cell membranes. HT-29, HEp-2, RK-13, and MDCK cells were slightly fixed with 0.25% paraformaldehyde (PFA) for 1 h and then were washed and treated with 37 µg/ml Pet for 30 min. After this treatment, cells were washed again and fixed properly with 4% PFA but not permeabilized. Nuclear DNA was stained with TO-PRO 3 (blue), Pet was detected using an anti-Pet polyclonal antibody followed by a secondary antibody anti-rabbit IgG coupled to rhodamine (red), and CK8 or CK10 (as appropriate) was detected using monoclonal anti-CK10 or anti-CK8 antibodies followed by a secondary anti-mouse IgG antibody coupled to fluorescein (green). The slides were observed by confocal microscopy.
