Palmitoylation site in Ykt6 is essential for yeast growth. Yeast strain SARY166 carrying temperature-sensitive mutant ykt6 transfected with empty vectors, wild-type Ykt6 vectors, and S¹⁹⁶C¹⁹⁷ mutant of Ykt6 vectors were plated on the selection media as described in Materials and Methods and incubated at 25°C or 37°C for 5 and 3 days, respectively. Two independent clones were shown in each transfectant. We confirmed that the expression levels of ykt6 at 25°C were comparable between wild-type and SC mutant transfectants in immunoblot analysis (data not shown).
