siRNA-mediated knockdown of RORα decreased CYP8B1 mRNA expression. A, HepG2 cells were transfected with RORα siRNA (180 and 300 ng) for 8 h to knockdown RORα expression. Scrambled siRNA to RORα (siControl 300 ng) was used as a negative control. Relative mRNA expression of CYP8B1, CYP7B1, and RORα was assayed by real-time PCR. B, Western blot analysis of RORα protein (10% acrylamide gel) using total HepG2 cell lysate transfected with RORα siRNA (180 and 300 ng) for 10 h. Scrambled siRNA (300 ng) (siControl) was used as a negative control. C, HepG2 cells were transfected with siRORα (180 and 300 ng) or scrambled siControl as a negative control for 4 h followed by treatment of 8-Br-cAMP (10 μm) for 8 h.
