Strength and specificity of DIG3-3 interaction with VirD2. (A) Yeast strains harboring the DIG3-3 prey were transformed with cVirD2, mcVirD2, flVirD2, or the A. nidulans GTPase baits and assayed for β-galactosidase activity as described in Materials and Methods. (B) In vitro interaction of DIG3-3 with VirD2. Purified T7-epitope tagged DIG3-3 was passed over glutathione-Sepharose columns that had bound GST, GST-cVirD2, or GST-flVirD2. Bound fractions were eluted and analyzed on Western blots with anti-T7 monoclonal antibodies. Lane 1, GST-cVirD2; lane 2, GST-flVirD2; lane 3, GST; lane 4, purified T7-tagged DIG3-3.
