PI3K inhibition down-regulates both AKT and ERK signaling in HER2 amplified and PIK3CA mutant breast cancer cells. (A) Cells were treated with 1 μM GDC-0941 for the indicated times, and lysates were probed with the indicated antibodies. Although BT474 has a K111N amino acid substitution, this mutation was found to have no effect on AKT phosphorylation (39). Independent experiments were performed three times, and a representative result is shown.
