(A) Each group of mice (n = 5) was injected by IM-EP with 50 µg of E7 DNA vaccines (pE7) plus 10 µg of IL-2 cDNAs (pIL-2) per mouse in a final volume of 50 µl at 0 and 1 weeks. The animals were also injected i.p. with 100 µg of anti-4-1BB and control rat Abs at 0 and 1 weeks. The animals were tested for in vivo CTL lytic activity at 2 weeks. For this test, E7 peptide-pulsed (CFSE high) and un-pulsed (CFSE low) splenocytes were injected i.v. into the immunized mice, as described in “Materials and Methods.” The mice were sacrificed after 8 h and the splenocytes were analyzed by FACS to measure the level of CFSE-labeled cells in each subset. M1, un-pulsed CFSE low population; M2, E7-pulsed CFSE high population.
