NA increases liver and blood NAD and NAM levels in vivo. PC3 tumor-bearing xenografts (n = 5 per group) were treated with vehicle, NA, GNE-617 or the combination of both agents for 5 days with the indicated doses and mouse liver (A and B) and blood (C and D) collected 1 hour following the final dose for measurement of NAD (A and C) and NAM (B and D) levels by LC-MS/MS as described in Materials and Methods section. Comparisons between NA-treated groups and control (vehicle) was determined by Dunnett method: ***P < .0001. Comparisons between treatment groups were performed using unpaired t test assuming unequal variances: #P < .01, ##P < .001.
