TWEAK binds to its receptor and activates an intracellular signalling cascade (*)12,23, which leads to phosphorylation of Keap1 or accordingly Nrf2 via various mitogen activated kinases27. This induces a disinhibition of Nrf2, which then represses NFkB expression as shown elsewhere (#)6,22. Its inhibition consequently leads to downregulation of TWEAK, as NFkB normally induces TWEAK expression (§)16.
