Histochemical analysis of intracellular ROS accumulation and ion leakage in leaves. (A) WT and osldc-like 1 HM leaves were incubated with H2O2 for 30 min. Leaf disc images were obtained by confocal microscopy after incubation with DCFH-DA for 10 min. In the case of treatment with spermine, leaf discs were co-treated with spermine in MES buffer containing 50 mM H2O2, 200 mM NaCl, or 10 mM potassium phosphate buffer (pH 3.0). (B) Ion leakage in H2O2-treated WT and osldc-like 1 rice plants. Ion leakage was also determined in H2O2-treated rice leaves co-treated with 1 mM spermidine. The leaves were incubated in the presence of 50 mM H2O2 for 48 h. Data are reported as the means ± SD.
